Some advice please if you book a family room for 4 and only 3 turn up will they charge you any more for under occupancy ?

Reason I ask is that the eldest has not decided whether to come away with us and we need a stopover as a long drive.

Is the correct answer provided the occupancy does not exceed the permitted number for that room. In your case your room may be of use from anything between 1 and 4 people.
